Selected data and study scope

This dataset contains snRNA-seq data from Macaca fascicularis, sampled from whole hypothalamus. The default comparison examines gene expression in Obese relative to Normal.

This obese-versus-normal reanalysis uses the macaque hypothalamic single-nucleus data produced by Lei and colleagues. It includes source samples HCA_Macaque001_S001 through S006. S001 through S003 are Normal and S004 through S006 are Obese. The analysis treats each cell population separately, so retained sample counts can differ between populations.

The explorer includes 60 cell populations and 1 comparison for this dataset.

The default track, e9 (POMC/TBX3), includes 3 Normal samples and 3 Obese samples in this comparison. These are analysis sample counts. Check the source for biological replication and pooling.

Interpretation and limitations

The source publication describes the original study. The obese-versus-normal reanalysis shown here is separate and does not yet have its own publication.

The source assay profiles nuclei. Check the population track and its retained sample count to see which analysis each result describes.

Expression values are CPM (TMM-normalized): counts per million on library sizes adjusted by the TMM factors of the study’s own edgeR fit. This scale adjusts for library size and composition, not gene length. Use it to inspect a feature across the samples in this analysis. CPM values are not absolute transcript counts.

Evidence and verification

Pipeline QC not assessed. macaque_snrna/obese_vs_normal; all 60 app tracks; saved outputs and a refit from the committed pseudobulk; nucleus-level processing remains unverified

Experimental design review needed. macaque_snrna/obese_vs_normal; all 60 app tracks; saved outputs and a refit from the committed pseudobulk; nucleus-level processing remains unverified

Author concordance not applicable. Outside author-validation scope

Pipeline QC can pass while experimental-design limitations remain. Read both assessments before interpreting results.

Open the dataset in the Explorer to read each check and its evidence. Author concordance compares effects for matched genes. It does not require identical author P values or FDR.

Obese vs Normal · default

Baseline: Normal → Case: Obese

Positive differential-expression values indicate higher expression in the case group relative to the baseline. Check the comparison direction and study design when interpreting results.
