Selected data and study scope

This dataset contains RiboTag translatome data from Mus musculus, sampled from whole hypothalamus. The default comparison examines gene expression in Aged (2 years) relative to Young (4 months).

The explorer includes 1 track and 1 comparison for this dataset.

The default track, Hypothalamic astrocytes (GFAP-RiboTag IP), includes 3 Young (4 months) samples and 3 Aged (2 years) samples in this comparison. These are analysis sample counts. Check the source for biological replication and pooling.

Interpretation and limitations

This preparation profiles ribosome-associated RNA from the targeted cells. Treat the result as a translatome measurement. It does not represent total RNA from every cell in the anatomical region.

Expression values are CPM (TMM-normalized): counts per million on library sizes adjusted by the TMM factors of the study’s own edgeR fit. This scale adjusts for library size and composition, not gene length. Use it to inspect a feature across the samples in this analysis. CPM values are not absolute transcript counts.

Evidence and verification

Pipeline QC passed. Mouse hypothalamic astrocytes — 2 years vs 4 months (RiboTag RNA-seq); all 1 registered contrasts on default track astrocytes

Experimental design passed. Mouse hypothalamic astrocytes — 2 years vs 4 months (RiboTag RNA-seq); all 1 registered contrasts on default track astrocytes

Author concordance passed. Selected author genes only; aged_vs_young. P values and FDR are contextual. Numerical acceptance: strong correlation and matched-gene direction.

Pipeline QC can pass while experimental-design limitations remain. Read both assessments before interpreting results.

Open the dataset in the Explorer to read each check and its evidence. Author concordance compares effects for matched genes. It does not require identical author P values or FDR.

Source study

Hypothalamic GFAP-RiboTag immunoprecipitate from 4-month-old vs 2-year-old mice, n=3/3 biological replicates. The single input library per age is excluded because it is not a replicated contrast (Boisvert et al., Cell Rep 2018, PMID 29298427; GSE99791)

Baseline: Young (4 months) → Case: Aged (2 years)

Positive differential-expression values indicate higher expression in the case group relative to the baseline. Check the comparison direction and study design when interpreting results.
